J'ai toujours utilisé dans mes séances la question : what's this?

Dans le manuel que j'ai, ils utilisent : what is it ?

Quelle est la différence et quelle formule vaut-il mieux utiliser avec les élèves?

Tous les 2 sont valables! L'avantage de "What's this?" est que ça introduit "this" (pour voir plus tard "that")

What's this se traduirait par "qu'est-ce que ceci ?"

Et What is it par "Qu'est-ce que c'est"

Les deux sont corrects. On dit plutôt What's this quand on désigne quelque chose.
